IZVLEČEK Geografska lega, klimatske razmere in zgodovinska pripadnost so glavni dejavniki, ki so botrovali k oblikovanju raznolikega sortimenta sort žlahtne vinske trte na območju današnje Slovenije. Sorte žlahtne vinske trte (Vitis vinifera L.) je mogoče opisati na več nivojih in z različnimi metodami. Ovrednotenje ali celo okarakterizacija lahko poteka na podlagi morfoloških, morfometričnih, biokemijskih in genetskih značilnosti posamezne sorte. V prispevku smo na podlagi analiz karotenoidnih pigmentov ter DNK analiz z uporabo molekulskih markerjev ugotavljali podobnosti in sorodnost med 17-imi akcesijami žlahtne vinske trte. Namen raziskave je bil odkriti morebitno povezavo med karotenoidnimi profili in meritvami barve sort ter ovrednotenje kemotaksonomskega potenciala dobljenih rezultatov za razlikovanje sort žlahtne vinske trte. Dobljene rezultate smo primerjali z rezultati analiz z metodo mikrosatelitov. V raziskavo smo vključili 17 različnih genskih virov belih sort žlahtne vinske trte, ki so posajene v kolekcijskem vinogradu Biotehniške fakultete (BF), ki obenem služi tudi kot genska banka. Rezultati evalvacije obravnavanih genskih virov belih sort žlahtne vinske trte se tako na biokemijskem kot tudi na genetskem nivoju dopolnjujejo, kar pomeni, da so obravnavane akcesije generalno razdelili v tri večje skupine, znotraj katerih prihaja do manjših odstopanj, kar je verjetno posledica lastnosti posameznih genotipov in vpliva okolja. Ključne besede: žlahtna vinska trta, genski viri, kemotaksonomija, karotenoidi, mikrosateliti, lokus ABSTRACT EVALUATION OF GENETIC RESOURCES OF WHITE GRAPEVINE VARIETIES (Vitis vinifera L.) Geographical location, climate and historical affinities are the main factors that have contributed to creating a diverse assortment of grapevine varieties in the area of Slovenia. The grapevine varieties (Vitis vinifera L.) can be described at several levels, using different methods. Evaluation can be done on the basis of morphological, morphometrical, biochemical and genetic characteristics of each variety. In this paper the biochemical analysis of carotenoid pigments and DNA analysis using molecular markers observed similarities between seventeen grapevine accessions. The purpose of this study was to find connections between carotenoids, shapes and measurements, evaluation of chemotaxonomical potential of obtained results to distinguish among varieties, and these properties compare with the results of the qualitative analysis with microsatellites. The study included 17 different genetic resources of white grapevine varieties, which are planted in the collection vineyard of Biotechnical Faculty (BF), which can also serve as a gene bank. The results of the evaluation with treated genotypes are supplemented on biochemical and genetic level. Generally, the accessions are divided into three major groups within which there are minor discrepancies, probably due to the properties of individual genotypes and the impact environment. Radojko PELENGIĆ, Barbara PIPAN, Vladimir MEGLIČ, Denis RUSJAN 1 UVOD Geografska lega, klimatske razmere in zgodovinska pripadnost so glavni dejavniki, ki so botrovali oblikovanju sortimenta sort žlahtne vinske trte na območju današnje Slovenije. S pojavom bolezni in škodljivcev na žlahtni vinski trti ob kocu 19. stoletja, ter zahteve po določenih tipih vina je prišlo do dodatne erozije genotipov pri žlahtni vinski trti, predvsem v škodo lokalnih in avtohtonih sort (Vertovc, 1845; Goethe, 1997; Pelengić in Rusjan, 2010). Sorte žlahtne vinske trte (Vitis vinifera L.) je mogoče opisati na več nivojih in z različnimi metodami. Evalvacija lahko poteka na podlagi morfoloških opisov različnih delov rastline pri različnih fenofazah, z morfometričnimi meritvami različnih rastlinskih delov, biokemijskih kvalitativnih ali kvantitativnih analiz in genetskih značilnostih posamezne sorte (Galet, 1979; O.I.V., 1983; Schneider in sod., 1988; Cabelo in sod., 1995; Martinez de Toda in sod., 1997; Asensio in sod., 2002; Kozjak in sod., 2003; Štajner in sod., 2008; Pelengić in Rusjan, 2010). V prispevku smo na podlagi analiz karotenoidnih pigmentov ter DNK analiz z uporabo molekulskih markerjev ugotavljali sorodnost med 17-imi akcesijami žlahtne vinske trte. Namen raziskave je bil dobiti morebitno povezavo med karotenoidnimi profili in meritvami barve, ob enem ovrednotenje kemotaksonomskih lastnosti, ki bi se lahko pokazale kot učinkovite za razlikovanje med sortami. Te meritve smo primerjali z rezultati analize mikrosatelitov. 2 MATERIAL IN METODE V raziskavo smo vključili 17 različnih genskih virov belih sort žlahtne vinske trte, ki so bili nabrani v kolekciji Ampelografski vrt, ki je del raziskovalnega vinograda Biotehniške fakultete (BF) in obenem služi tudi kot genska banka. Nahaja se v Kromberku pri Novi Gorici. Skrbnik vinograda je BF, Oddelek za agronomijo, Katedra za hortikulturo. Vzorčenje mladih listov je potekalo v začetku rastne dobe in je vključevalo naslednje genske vire: 'Bela Glera (5-6)', 'Glera (I)', 'Zelen Goče', 'Zelen Kolekcija', 'Beli Teran', 'Glera (II)', 'Dišečka', 'Pokov Zelen', 'Vitovska', 'Dišeči Traminec', 'Sauvignon blanc', 'Racuk A1-3', 'Rumeni Muškat', 'Medena Glera', 'Bela Glera (8-1)', 'Verduc' in 'Rebula'. Pri vseh akcesijah smo s tekočinsko kromatografijo visoke ločljivosti (High Performance Liquid Chromatography-HPLC) izmerili vsebnost violaksantina, neoksantina, anteraksantina, luteina, β-karotena, klorofila a in klorofila b, ter s kolorimetrom barvo jagod po sistemu CIELAB. Vse sorte smo vključili tudi v genetsko analizo z enajstimi specifičnimi mikrosatelitnimi markerji (VVMD5, VVMD7, VVMD25, VVMD27, VVMD36, VrZAG21, VrZAG47, VrZAG62, VrZAG79, VrZAG83, in VrZAG112) (Sefc in sod.,????). 3 REZULTATI Z DISKUSIJO Karotenoidni profili se pri preučevanih akcesijah med seboj razlikujejo. Sorti 'Zelen Goče' in 'Bela Glera (8-1)' imata večjo vsebnost violaksantina, medtem ko sorte zapisane kot 'Bela Glera (5-6)', 'Glera (I)', 'Verduc', 'Zelen Kolekcija', 'Pokov Zelen', 'Bela Glera (8-1)' in 'Zelen Goče' imajo večjo vsebnost β-karotena in luteina. Povprečna vsebnost ksantofilov je 5,48 µg/g, luteina 2,27 µg/g, violaksantina 1,78 µg/g, neoksanthina in 1,06 µg/g anteraksantina. Ovrednotenje genskih virov belih sort žlahtne vinske trte (Vitis vinifera L.) bila izmerjena pri sorti 'Bela Glera (8-1)', najmanjša (3,41 µg/) pa pri sorti 'Vitovska'. Največjo vsebnost klorofilov je imela sorta 'Pokov Zelen' (134,2 µg/g) in najmanjšo pa sorta 'Vitovska' (30,4 µg/g). V tej fazi projekta smo na osnovi molekularnih analiz ugotovili, da na lokusih VVMD5, VVMD36, VrZAG79 in VrZAG112 ni razlik med posameznimi akcesijami. Amplifikacija lokusov VVMD25, VVMD27 in VrZAG27 je bila uspešna le pri sorti 'Bela Glera (8-1)', lokus VVMD7 se je uspešno namnožil pri sorti 'Vitovska'. Največ razlik med akcesijami so se pokazale na lokusih VrZAG21 in VrZAG83. Prvi se je uspešno namnožil pri sortah 'Bela Glera (8-1)', 'Dišečka', 'Glera (I)', 'Rebula', 'Verduc' in 'Vitovska', lokus VrZAG83 pa se je pojavljal pri sortah 'Bela Glera (5-6)', 'Glera (I)', 'Racuk', 'Rumeni Muškat' in 'Vitovska'. Pri sortah 'Beli Teran', 'Vitovska' in 'Verduc' je bil uspešno namnožen lokus VrZAG62.
